Design and Usability

The NucleoCounter ® NC-3000™ is a state-of-the-art cytometer that uses fluorescence imaging to characterize cell properties.

The NC-3000™ can perform cell counting and viability determination at high speed and includes plug-and-play analytical assays for mammalian, yeast and insect cells. Predefined assays include a single 5-minute cell cycle test with a corresponding selection of apoptosis assays.

With the FlexiCyte™ license, the user can create protocols by choosing the light source, emission filters and detailed settings. The accompanying intuitive software can be used for data analysis of the results and has unlimited licenses.

The NucleoCounter® NC-3000™ advanced imaging cytometer ensures flow analysis with high reproducibility and no calibration required..

The scientist can count the cells manually, visually verifying the count. This verification can also be performed with the intuitive NucleoView™ software included with the NC-3000™. Scatter plots and images are interrelated, allowing visual verification of cell populations.

NucleoView™ software allows easy coupling between the image obtained and the graphs showing fluorescence for accurate determination of the events to be counted.

The closed cell counting system, which uses self-contained disposable cassettes, helps the user to be protected and to be when handling samples. The absence of pipettes, the exposure to (toxic) reagents and minimal cleaning make the NucleoCounter® a safe choice. NucleoCounter® a safe choice.

The NucleoCounter® NC-3000™ can count cells even from a sample of highly aggregated cells or cells growing in clusters.

In the Aggregate Cell Assay, the total cell count is obtained from a lysed aliquot of the sample (see schematic below).

Lysis disrupts the cell membrane, dismantles the clumps and releases the suspended nuclei. Nuclei are stained with DAPI and counted automatically. Since dead cells are usually less abundant and tend to detach from the aggregates, the dead cell count is determined from the unlysed aliquot (figure below).

The combination of these two measurements results in high precision and accuracy even for the most difficult groups.

Cell cycle analysis is the fundamental method for establishing abnormalities in cell division and, therefore, symptoms of cell damage. With ChemoMetec’s unique 2-step cell cycle, a cycle can be performed in 5 minutes including cell treatment.

Its profile will be displayed in the accompanying PlotManager software and the percentage or number of events in G0/G1, S-phase, G2 and sub-G1 can be easily identified.

With the NucleoCounter® NC-3000™ plug-and-play GFP transfection assay, the number of viable and dead cells transfected with GFP can be determined with high accuracy.

The NucleoCounter-3000™ is ideal for regulated quality control, production and even R&D and PD departments. The instrument and accompanying software can be easily integrated into existing computer networks.

The FlexiCyte™ module enables users to perform advanced cellular analysis of a wide range of biomarkers and fluorescent proteins in mammalian cells.
