The NucleoCounter® SP-100™ Sperm Cell Counter offers a unique ease of use for the effective determination of total sperm concentration and sperm viability in an ejaculate or extended dose semen sample.

The NucleoCounter® SP-100™ is very easy to operate and measurements can be performed by operators with minimal technical training. It is a compact instrument that fits perfectly in any research, quality control or production site.

The measuring range of the NucleoCounter® SP-100™ is virtually unlimited due to the pre-analysis dilution.

Determination of cell viability in sperm cell samples

The NucleoCounter® SP-100™ concept offers the determination of cell viability in addition to the total cell count in the sperm sample. The determination of viability is based on the “PI staining” method.

Measuring sperm cell viability

Cell Viability (membrane integrity) can be measured with the NucleoCounter® SP-100™ in a two-step procedure by measuring the difference between the total cell count and the non-viable cell count (see figure below).

When a sample is diluted with S100 Reagent, lysis of the viable cell membrane occurs, rendering all cell nuclei susceptible to propidium iodide (PI) staining, resulting in a total cell count.

However, nonviable cells diluted with PBS (phosphate buffered saline) are permeabilized without treatment and therefore stain directly with PI, resulting in a nonviable cell count. The difference between the two measurements gives the viable cell count – or % viability.
