Selective breeding by artificial insemination is widely used to improve the characteristics of dairy cows, beef cows, pigs and horses.

Sperm count estimation using a photometer is an old method that is still widely used. However, the photometer method is an indirect method of measurement and is prone to interference from, for example:

  • Dust
  • Waste
  • Gel particles

The NucleoCounter® SP-100™ is a unique cell counter that maximizes the credibility of sperm counts and can provide direct economic benefits. As the coefficient of variance is less than 3%, fewer cells are needed to ensure a specific minimum number present in 95% of the samples. The use of a diluent does not interfere with the high accuracy of the SP-100™ as the detection method is direct, using propidium iodide DNA staining.

Types of Samples:

Selective breeding by artificial insemination is widely used to improve the characteristics of pigs. This procedure favours the transmission of reproductive traits, but also influences:

  • Structural correction
  • Size, type and muscle allocation
  • The fat cover

To maximize the chances of a full litter it is important to have a sufficient number of sperm when inseminating. This is usually 80 ml containing between 2.3 and 2.5 billion cells in Europe, and more than 3 billion cells in America.

On average, a boar ejaculate produces between 10 and 100 billion sperm. If each dose is packaged so that there are a minimum of 3 billion cells in 95% of the sample on average, then the SP-100™ will provide up to 31 doses, whereas working with a photometer will only result in 26-27.

The SP-100™ can also determine sperm viability. A boar can have a large semen production, but with a very low quality because many of the sperm are dead. Instead of wasting resources on seemingly high quality boars with low viability sperm, one can focus on the animals with the highest viability and maximize the chances of a full litter by using viable sperm count instead of total sperm count.

When inseminating a cow it is very important to have a certain number of cells to achieve the best chance of successful impregnation.

On average, a bull ejaculation produces about 5 billion sperm. If each sheath contains 30 million cells, the NucleoCounter® SP-100™ will provide 150-160 sheaths, whereas working with a photometer will only provide 130.

When inseminating a mare it is very important to have a certain number of cells available to maximize the chances of successful insemination.

After obtaining the ejaculate, a diluent is added which contains sperm sustenance and counteracts the effects of the seminal fluid that transports the sperm during ejaculation. If a diluent is not added, the seminal fluid can kill the sperm.

Semen from stallions varies greatly in how well it freezes and which specific extender gives the best results. With the NucleoCounter® SP-100™ you will be able to find the best choice as it provides not only the total sperm count but also the percentage of live cells. Dividing a semen sample into multiple aliquots and freezing with different diluents, followed by viability determination, provides fast and accurate results that can increase the success rate of insemination.
